In several animal species, ageing process usually leads to alterations in plasticity and in synaptic transmission in different brain regions, and correlates with alterations in memory and learning. Our group of investigation has a vast experience in the study of the Sprague-Dawley senile female rat as a model of spontaneous ageing with age.
One of the aims of the present line is to characterize the hippocampus and the brain structures related to senile rat (28 months of age) and middle-aged rat (12 months of age), which show age-related cognitive deficit. The characterization is carried out from the cognitive, histopathological and molecular level.
Cognitive function is evaluated using the Barnes maze, which determines the assessment of spatial memory, in addition to other methods to determine spatial discrimination and characteristics of objects (SOR and NOR, respectively).
In histopathology, immunomarking techniques are applied (immunohistochemistry and immunofluorescence), bright field microscopy, fluorescence, and confocal microscopy with stereology-based morphometric methods.
At the molecular level, gene expression is mainly evaluated by PCR or RNAseq.
Another objective is to imitate or reinforce the protective mechanisms which are found diminished or depleted in brain ageing. As therapeutic strategy we use gene therapy, and as biotechnological tool we have built adenoviral and lentiviral vectors carrying genes such as IGF-I, GDNF, TYROBP and Yamanaka genes (Sox2, c-Myc, Klf4 and Oct4).
